My gas fire has recently started making a chuffing or puffing sound. It is intermittent but happens every few seconds and is accompanied by a slight change in the flame. The appliance is 15-20 years old and has a glass front and fake coals.

Any ideas as to what could be causing this, and whether it needs correcting?

Get the appliance serviced. The engineer will correct any problems while he is there.
